Oric Pharmaceuticals Inc (NASDAQ:ORIC) saw a modest bump early Friday, with shares up roughly 1.9% in premarket, following the release of a peer-reviewed paper focusing on its lead cancer candidate, enozertinib.
The study, published in the journal Cancer Research, sheds light on enozertinib's journey from discovery to development. This compound selectively targets EGFR exon 20 mutations prevalent in non-small cell lung cancer (NSCLC), notable for its ability to cross the blood-brain barrier - a key consideration given the prevalence of brain metastases in lung cancer patients.
Preclinical trials showcased enozertinib's strong kinase selectivity, potent anti-tumor activity, and significant brain penetration. A highlight was a documented case of a patient exhibiting complete remission of both systemic tumors and brain metastases attributed to an EGFR exon 20 insertion mutation, which Oric points to as a unique achievement among similar drugs.
Melissa Junttila, Oric's vice president and head of biology, conveyed optimism that enozertinib stands apart in addressing the challenges faced by patients with these specific mutation-driven lung cancers. The focus on both systemic and central nervous system responses could mark a meaningful advancement in treatment options.
Looking ahead, Oric plans to unveil further clinical findings at the ESMO Asia 2025 conference this December. These updates will cover data from both first-line and second-line treatment cohorts using enozertinib for EGFR exon 20 mutations. Additional results expected by mid-2026 could include first-line atypical mutation treatments and combination therapies involving its collaborator's amivantamab.
While the market reaction has been positive so far, the stakes remain high. Oncology drug development is littered with promising compounds that stumble in later trials. Yet, the published data and the patient case offer something concrete for the scientific and investor community to chew on.
It's worth noting that the drug class targeting EGFR exon 20 mutations has been historically challenging for researchers, partly due to the intricate nature of these mutations and difficulties in reaching sanctuary sites like the brain. Enozertinib's reported profile, especially its brain penetration capability, ticks several critical boxes that have eluded other entrants.
