Shares of Rezolute, Inc. (NASDAQ:RZLT) took a severe hit, plummeting about 90% after announcing disappointing top-line results from their Phase 3 sunRIZE study. This trial assessed ersodetug in patients with congenital hyperinsulinism but missed the mark on both the primary and key secondary endpoints.

The company revealed that the highest tested dose of ersodetug (10 mg/kg) led to roughly a 45% decrease in hypoglycemia events, yet this reduction did not reach statistical significance when compared to a 40% improvement experienced by the placebo group. Similarly, the secondary measure - analyzing average daily time spent in hypoglycemia - showed a 25% reduction for the treatment arm versus a 5% increase for placebo, which also fell short of statistical significance.

Brian Roberts, M.D., Rezolute's Chief Medical Officer, expressed the company's disappointment, recognizing the urgent need for new therapies in this rare disease space. Despite the trial's failure to demonstrate clear glucose control benefits, the safety profile appeared generally favorable, though two subjects experienced serious hypersensitivity reactions leading to early withdrawal. The most commonly reported side effect was mild, temporary hypertrichosis.

Rezolute plans to engage with the FDA utilizing its Breakthrough Therapy Designation to explore future avenues for this program. Meanwhile, the firm's ongoing Phase 3 upLIFT trial for tumor-induced hyperinsulinism moves ahead, with readouts anticipated in the latter half of 2026.
